'Nicotinamide riboside is one of the better-studied NAD+ precursors, and the evidence consistently shows that it raises NAD+ levels in humans,' says Gary Soffer, M.D., pediatric allergy and immunology physician at Yale Medicine and medical director of Smilow Cancer Hospital at Yale New Haven Health. 'The challenge is that increasing NAD+ has not yet been shown to have meaningful improvements in health, longevity or physical function.'
Baljash S. Cheema, M.D., cardiologist and medical director of the Human Longevity Clinic at Northwestern Medicine, says that while it's 'biologically plausible' that supplements that can boost NAD+ levels may be effective for longevity purposes, there isn't enough evidence to suggest that taking an oral supplement will improve your health.
'We have to stay grounded in what the science shows,' says Dr. Cheema, adding that there have been positive findings in animal studies but that, generally speaking, positive results from pre-clinical trials (including animal studies) don't necessarily mean those results will be replicated in human studies.
The science on NR supplementation in humans is relatively new (within the last decade) and includes mixed results, making it difficult to decipher benefits other than the fact that NR supplements do increase NAD+ levels. Authors of a 2025 review in Clinical Bioenergetics, which combed over the available research on NR and NMN supplements, concluded the benefits 'appear to be most pronounced in individuals with suboptimal baseline NAD+ levels,' and that 'NR use may offer little advantage beyond increasing blood NAD+ concentrations'.
