Think of a drug as a key and the protein it binds to as a lock. Roughly a third of approved drugs are aimed at one particular kind of lock: G protein-coupled receptors (GPCRs). The problem is that this lock doesn't hold still — it keeps shifting shape, so it's often unclear which shape you'd even need to cut the key for. That's why drug discovery has long treated GPCRs as some of its hardest targets. One company betting that AI and structural biology can crack that problem just raised a large new round. Where Computational Drug Discovery Met Biology Superluminal Medicines is a Boston-based biotech company integrating AI/ML, protein dynamics and structural biology to target GPCRs. Its 'Agentic Cryo-EM' capability uses cryo-electron microscopy — which flash-freezes proteins and images their actual 3D shape, essentially photographing them — and runs that process through AI agents that repeat it at scale without human intervention, generating hundreds of empirical GPCR structures. The company combines those structures with 'co-folding' models that predict exactly how a candidate molecule locks onto a protein, de novo molecule design tuned to each of the many shapes a binding pocket can take, and predictive models for how a drug would be absorbed, distributed, metabolized, excreted and how toxic it might be — all to speed up finding viable drug candidates. Its lead program is a selective, biased MC4R agonist for rare genetic forms of obesity and hypothalamic obesity, expected to enter Phase 1 trials by the end of this year. The MC4R pathway is a clinically validated regulator of appetite and energy metabolism, with particular relevance to rare genetic obesity conditions such as Bardet-Biedl syndrome. Superluminal says its approach activates only the signaling pathways responsible for the desired effect, minimizing side effects. Beyond its own pipeline, the company also has an active GPCR-focused collaboration with Eli Lilly and Company targeting cardiometabolic diseases and obesity. CEO Cony D'Cruz spent eight years as Executive VP and Chief Business Officer at computational drug discovery firm Schrödinger, and previously served as President of Proteros and SVP of North American business development at Evotec. Co-founder and Head of Discovery Ajay Yekkirala was a postdoc at Harvard Medical School and Boston Children's Hospital and co-founded Blue Therapeutics before serving as VP, Head of Biology at RA Capital Management. The two co-founded Superluminal in 2023. Superluminal announced an oversubscribed $60 million Series B led by BVF Partners. New investors Deep Track Capital and Perceptive Advisors joined, alongside existing backers RA Capital Management, Insight Partners, NVIDIA, Catalio Capital Management, Eli Lilly and Company, Cooley and Gaingels. The company previously raised a $33 million seed round in August 2023 and a $120 million Series A in September 2024, both led by RA Capital Management — a firm both co-founders have direct ties to. Harsha Paladugu, a BVF board member, said the lead MC4R program addresses a serious unmet need for patients with rare genetic forms of obesity, and that Superluminal's integrated discovery platform has shown it can generate differentiated small molecules against historically difficult GPCR targets. Nandita Shangari, a managing director at RA Capital Management and board member, said the caliber of the investor syndicate itself reflects the progress the company has made. Others Racing to Crack the Same Problem Superluminal isn't the only company trying to accelerate drug discovery with AI, and different players attack different links in the same chain. Xaira Therapeutics, co-founded by Nobel laureate David Baker, launched with $1 billion on day one — a step earlier in the pipeline than Superluminal, using protein design models like RFdiffusion to build entirely new molecules and antibodies from scratch. Isomorphic Labs, an Alphabet company, has extended DeepMind's AlphaFold technology beyond proteins to DNA, RNA and small-molecule interactions — and, like Superluminal, has its own partnership with Eli Lilly (as well as a separate one with Novartis). Genesis Molecular AI has raised more than $300 million around Pearl, a foundation model the company says outperforms AlphaFold 3 on key benchmarks. Iambic Therapeutics raised a $100 million Series B combining physics-based simulation with data-driven prediction, and has its own partnership with Nvidia.
